“Where did the drug actually go?” That one question can determine whether a clinical candidate advances or gets shut down. In a packed session, Matt Silva, PhD (CEO of Emit Imaging), introduced one of the most eye-opening technologies I’ve seen in a while: Cryo-Fluorescence Tomography (CFT) — a high-res, 3D imaging method that allows you to map gene or cell therapies across an entire animal with voxel-level clarity. Imagine: ✔️ Seeing where your AAV vector actually expresses (not just assuming based on IHC or PCR) ✔️ Spotting unexpected signals in the jaw, tongue, or marrow before they surprise regulators ✔️ Screening out poor candidates early, based on whole-body distribution patterns ✔️ Multiplexing different fluorophores to evaluate delivery, expression, and clearance in one run And all of this is ex vivo — integrating seamlessly with your existing workflow (tissue banking, histology, mass spec, etc.). This isn't just pretty imaging. It’s a decision-making engine. CFT helped kill a candidate with unexpected marrow uptake. It revealed sub-organ signals in lymph nodes, pancreas, and even dental roots. This is not something you'd casually catch on 2D images or bulk PCR. I recently sat in on a talk from Dr. Jason DelCarpini, Director of Bioanalytical and Molecular Assays at Moderna, and it was quite the lesson in clarity, leadership, and real-world problem-solving. Jason broke down a key issue in our field: inconsistent neutralizing antibody (NAb) reporting across ligand-binding and cell-based assays is creating confusion, review delays, and missed opportunities to streamline regulatory approval. So what’s the fix? The BAR Template—developed through an industry-led white paper Jason co-authored—is a proposed standard for NAb sample testing and reporting. It offers: ✔ Clear section-by-section guidance ✔ Transparent reporting of methods, controls, deviations, and analysis ✔ A common language regulators can quickly follow Instead of every lab reinventing the wheel, this framework promotes harmonization across submissions—something we desperately need with increasingly complex biologics and multi-domain therapies. What a time we had for dinner and scientific discussion at Ocean Prime on Monday at AAPS NBC!! Johannes Stanta led an interactive discussion on how bioanalytical methods must adapt as cell-based, organ-on-chip, and tissue models begin to replace traditional GLP safety testing, following new FDA guidance. A timely topic with a rich discussion across the group. It mixed well with some great scientific networking and delicious food! We visited old friends and made some new ones and came out a bit wiser on what's next in GLP safety testing.
